    Offshore oil & gas installations in the Arctic: responding to uncertainty through science and law

    The Arctic Ocean’s physical environments and ecosystems are some of the most fragile and least well understood on Earth. They are characterised by extreme light and dark cycles, shortened food chains, and slow ecosystem recovery from disturbance. The Arctic seabed also holds promise of lucrative oil and gas resources, whose future exploitation could have substantial environmental impacts. Arctic jurisdictions must weigh environmental conservation and global agreements to reduce carbon emissions against the social implications and potential economic gain of offshore oil and gas projects in the Arctic, and must do so in the face of substantial scientific uncertainty around the impacts of climate and environmental change in the Arctic. We know, however, that major projects such as oil and gas projects have the potential to lead to transboundary environmental harm. We have some understanding of how any pollution may be carried by sea ice or on the ocean currents which flow around the Arctic Ocean. Even so, we have little understanding of how such pollutants might affect the Arctic ecosystem. Substantial gaps remain in scientific understanding of Arctic ecosystem functioning, particularly as it changes rapidly with the advent of climate change. These gaps in scientific understanding raise legal questions about how, for example, the law’s obligation not to cause significant transboundary environmental harm applies in the Arctic. In particular one may ask what actions are required by a state to show that they have acted with due diligence. Is it sufficient, for example, to show that they have complied with existing international treaties? This paper draws out key legal and scientific issues on which greater understanding is required. In essence it presents a roadmap for further research and negotiation

    Should I stay or should I go? Modelling year-round habitat suitability for fin whales in the California Current

    Aim Understanding the spatial ecology of endangered species is crucial to predicting habitat use at scales relevant to conservation and management. Here, we aim to model the influence of biophysical conditions on habitat suitability for endangered fin whales Balaenoptera physalus, with a view to informing management in a heavily impacted ocean region. Location We satellite-tracked the movements of 67 fin whales through the California Current System (CCS), a dynamic eastern boundary upwelling ecosystem in the Northeast Pacific. Methods We use a multi-scale modelling framework to elucidate biophysical influences on habitat suitability for fin whales in the CCS. Using Generalised Additive Mixed Models, we quantify the influence of a suite of remotely-sensed variables on broad-scale patterns of occupancy, and present the first year-round, high-resolution predictions of seasonal habitat suitability. Further, we model the influence of contemporaneous biophysical conditions on individual-level residence times in high-use habitat. Results We present evidence of year-round habitat suitability in the southern California Current System, robust to inter-annual variability, establishing that North Pacific fin whales do not follow the canonical baleen whale migration model. Within the high-use habitat in the Southern California Bight (SCB), individual-level residency to localised areas (n=16 for >30 days; n=4 for >6 months) was associated with warm, shallow, nearshore waters (>18°C, <500m); with cool waters (14-15°C) occurring over complex seafloor topographies and convergent (sub-)mesoscale structures at the surface. Main Conclusions Biophysical conditions in the southern CCS generate productive foraging habitats that can support the fin whale population year-round and allow for extended periods of residency in localised areas. High-use habitats for fin whales are co-located with areas of intense human use, including international shipping routes and a major naval training range. Seasonal habitat suitability maps presented here could inform the management of anthropogenic threats to an endangered baleen whales in this globally significant biodiversity hotspot

    Multiple uncontrolled conditions and blood pressure medication intensification: an observational study

    Abstract Background Multiple uncontrolled medical conditions may act as competing demands for clinical decision making. We hypothesized that multiple uncontrolled cardiovascular risk factors would decrease blood pressure (BP) medication intensification among uncontrolled hypertensive patients. Methods We observed 946 encounters at two VA primary care clinics from May through August 2006. After each encounter, clinicians recorded BP medication intensification (BP medication was added or titrated). Demographic, clinical, and laboratory information were collected from the medical record. We examined BP medication intensification by presence and control of diabetes and/or hyperlipidemia. 'Uncontrolled' was defined as hemoglobin A1c &#8805; for diabetes, BP &#8805; 140/90 mmHg (&#8805; 130/80 mmHg if diabetes present) for hypertension, and low density lipoprotein cholesterol (LDL-c) &#8805; 130 mg/dl (&#8805; 100 mg/dl if diabetes present) for hyperlipidemia. Hierarchical regression models accounted for patient clustering and adjusted medication intensification for age, systolic BP, and number of medications. Results Among 387 patients with uncontrolled hypertension, 51.4% had diabetes (25.3% were uncontrolled) and 73.4% had hyperlipidemia (22.7% were uncontrolled). The BP medication intensification rate was 34.9% overall, but higher in individuals with uncontrolled diabetes and uncontrolled hyperlipidemia: 52.8% overall and 70.6% if systolic BP &#8805; 10 mmHg above goal. Intensification rates were lowest if diabetes or hyperlipidemia were controlled, lower than if diabetes or hyperlipidemia were not present. Multivariable adjustment yielded similar results. Conclusions The presence of uncontrolled diabetes and hyperlipidemia was associated with more guideline-concordant hypertension care, particularly if BP was far from goal.     Production of Androgens by Microbial Transformation of Progesterone in Vitro: A Model for Androgen Production in Rivers Receiving Paper Mill Effluent

    We have previously documented the presence of progesterone and androstenedione in the water column and bottom sediments of the Fenholloway River, Taylor County, Florida. This river receives paper mill effluent and contains masculinized female mosquitofish. We hypothesized that plant sterols (e.g., β-sitosterol) derived from the pulping of pine trees are transformed by bacteria into progesterone and subsequently into 17α-hydroxyprogesterone, androstenedione, and other androgens. In this study, we demonstrate that these same androgens can be produced in vitro from the bacterium Mycobacterium smegmatis. In a second part to this study, we reextracted and reanalyzed the sediment from the Fenholloway River and verified the presence of androstadienedione, a Δ1 steroid with androgen activity

    Combinatorial multivalent interactions drive cooperative assembly of the COPII coat

    Protein secretion is initiated at the endoplasmic reticulum by the COPII coat, which self-assembles to form vesicles. Here, we examine the mechanisms by which a cargo-bound inner coat layer recruits and is organized by an outer scaffolding layer to drive local assembly of a stable structure rigid enough to enforce membrane curvature. An intrinsically disordered region in the outer coat protein, Sec31, drives binding with an inner coat layer via multiple distinct interfaces, including a newly defined charge-based interaction. These interfaces combinatorially reinforce each other, suggesting coat oligomerization is driven by the cumulative effects of multivalent interactions. The Sec31 disordered region could be replaced by evolutionarily distant sequences, suggesting plasticity in the binding interfaces. Such a multimodal assembly platform provides an explanation for how cells build a powerful yet transient scaffold to direct vesicle traffic
